摘要

目的:观察葛根汤、桂枝汤对颈椎间盘组织白介素1β（IL-1β）、诱导型一氧化氮合酶（iNOS）mRNA表达的调节作用。方法:选择8月龄雄性新西兰白兔36只

随机平均分为正常对照组、风寒湿刺激组、低头位组、风寒湿加低头位组、葛根汤组和桂枝汤组。通过建立风寒湿痹证型、低头位、风寒湿加低头位颈椎间盘退变家兔模型

采用RT-PCR技术检测颈椎间盘细胞因子IL-1β、iNOS mRNA的表达情况。结果:各个模型组与对照组比较

IL-1β、iNOS mRNA的表达上调（P<0.01）

葛根汤、桂枝汤组与风寒湿刺激组比较

IL-1β、iNOS mRNA表达均下调

有显著性差异（P<0.05或P<0.01）。结论:椎间盘退变中IL-1β、iNOS mRNA的表达上调

葛根汤下调IL-1β、iNOS mRNA表达

桂枝汤调节作用低于葛根汤。

Abstract

Objective: To investigate regulative actions of Gegen Decoction

Guizhi Decoction on expressions of interleukin-1β（IL-1β） and inducible nitric oxide synthase （iNOS） mRNA in the cervical intervertebral disc. Methods: 36 male New Zealand rabbits

aged 8 months

were randomly divided into a normal group

wind-cold-damp stimulation group

low head position group

wind-cold-damp plus low head position group

Gegen Decoction group and Guizhi Decoction group

and the cervical intervertebral disc retrograde degeneration rabbit models of wind-cold-damp arthralgia-syndrome type

low head position

wind-cold-damp plus low head position were established. Expressions of IL-1β and iNOS mRNA in cervical intervertebral disc were detected by RT-PCR technique. Results: The expressions of IL-1β and iNOS mRNA in the all model groups were up-regulated as compared with the control group （P<O.01）; the expressions in the Gegen decoction and the Guizhi Decoction groups were down-regulated as compared with the wind-cold-damp stimulation group（P<0.05 or P<0.01）. Conclusion: Expressions of IL-1β and iNOS mRNA in the cervical intervertebral disc of retrograde degeneration are up-regulated

and Gegen Decoction can down-regulate the expressions of IL-1β and iNOS mRNA

and the regulative action of Guizhi Decoction is lower than that of Gen Gen Decoction.
