摘要

目的:从分子生物学角度探讨 APOE 基因多态性与肾精亏虚型老年痴呆发病的相关性。方法:将41例老年痴呆患者及85例健康对照老年人分别按中医辨证分为肾精亏虚证、痰浊蒙窍证、瘀血阻络证。用 NaI 分离白细胞中的 DNA

multi-ARMsPCR 检测 APOE 基因型

并作出相关性分析。结果:（1）痴呆组基因型ε3/ε3与对照组比较差异有显著性（P<0.05）

痴呆组ε4/ε4和等位基因ε3、ε4与对照组比较也有显著性差异（P<0.01）。（2）ε4等位基因与老年痴呆发病的相对危险度（OR 值）为3.92>1。（3）肾精亏虚证与ε4等位基因有显著的相关性（P<0.01）。结论:（1）ε4等位基因为老年痴呆的危险因子

而ε3等位基因为老年痴呆的保护因子。（2）肾精亏虚证与ε4等位基因密切相关。

Abstract

Objective:To explore correlativity of senile dementia of deficiency of kidney essence with polymorphophism of APOE gene. Methods:41 cases of senile dementia and 85 healthy aging persons were divided into 3 TCM syndrome types:syndrome of deficiency of kidney essence

syndrome of orifices confused by phlegm

and syndrome of obstruction of blood stasis in collaterals. DNA in white blood cells was isolated by NaI

and APOE gene was detected by milti-ARMs PCR

and then correlative analysis was made.Results:（1）There was a significant difference between the dementia group and the control group in the gene type ε3/ ε3（P<0.05）

and ε4/ε4 and allelic genes ε3 and ε4（P<0.01）.（2）The relative risk（OR）of ε4 allelic gene with senile demendia attack was 3.92>1.（3）There was significant correlativity between syndrome of deficiency of kidney essence and APOE ε4 allelic gene（P<0.01）.Conclusion:（1）APOE ε4 allelic gene is risk factor and APOE ε3 allelic gene is protective factor for senile dementia.（2）There is close correlation between syndrome of deficiency of kidney essence and APOE ε4 allelic gene.
