Experimental Study on Concerted Application of Complex Prescription for Reducing the Renal Toxicity of Aristolochia manshuriensis Kom
Objective:To observe acute and chronic renal toxicity of the traditional prescription Daochi San containing Aristolochia manshuriensis Kom(GMT) in rats
and to probe into correlativity of concerted application
dose
administration time of the prescription with renal intoxicity of GMT.Methods:The rats were administrated with Decoction of GMT and Daochi San by intragastric infusion. The dosage of GMT is 6g/kg
12g/kg and 24g/kg each day for 7 days (acute toxic experiment)
and 4g/kg each day for 12 weeks(chronic toxic experiment)
respectively.Changes of renal function and morphology of the renal tissue were investigated in all groups;Dissolved amount of aristolochic acid A in the Decoctions of single GMT
Daochi San and separated drugs of Daochi San were detected with high performance liquid chromatography. Results:All different doses of GMT induced injuries of renal function and renal morphology in rats
which were positively correlated with the dose and administration time.Concerted application of compound Daochi San could remit the injury of single GMT on renal function and renal morphology
and the renal toxicity was significantly decreased by rational dose and administration time. The dissolved amount of aristolochic acid A in compound Daochi San Decoction was lower than that in single Decoction. Conclusion:Complex prescription Daochi San can decrease renal toxicity of GMT
and Daochi San and/or bamboo leaves have significant action of decreasing the dissolved amount of aristolochic acid A in GMT.
