摘要

目的探讨益气活血方对心肌梗死后大鼠缺血心肌局部血管新生的影响及其分子学机制。方法建立心肌梗死模型后

将大鼠分为5组

即益气活血方大剂量组（简称大剂量组）、益气活血方小剂量组（简称小剂量组）、麝香保心丸组、假手术组、模型组。灌胃6周后处死动物

取心肌组织

用免疫组化法测定梗死边缘区新生血管数（MVC）及微血管面密度（MVD）;检测缺血心肌局部胰岛素样生长因子-1（IGF-1）的表达。结果与假手术组相比

模型组大鼠心肌梗死边缘区MVC、MVD及IGF-1的表达明显升高（P<0.05）。大剂量组梗死边缘区MVC、MVD明显升高

与其他各组（除小剂量组外）比较差异均有显著性意义（P<0.05）;小剂量组有一定的促血管新生效应

其MVC较模型组差异有显著性意义（P<0.05）

MVD虽高于模型组

但两者比较差异无显著性意义（P>0.05）。大、小剂量组的IGF-1表达均高于模型组（P<0.05）

大剂量组与麝香保心丸组比较

差异有显著性意义（P<0.05）;小剂量组与麝香保心丸组比较差异无显著性意义（P>0.05）。结论益气活血方大剂量组能够促进梗死后大鼠缺血心肌局部血管新生

对缺血心肌具有保护作用

上调心肌局部IGF-1蛋白表达可能是其作用机制之一。

Abstract

Objective To study the molecular mechanism and effect of Yiqi Huoxue Formula（YHF） on angiogenesis in ischemic myocardium of rats after myocardial infarction.Methods Myocardial infarction model rats were divided into large dose YHF group

small dose YHF group

Shexiang Baoxin Pill group

sham operation group and model group.Rats were executed and myocardium was taken after 6 weeks’ intragastric administration.Microvessel counting（MVC）

microvessel density （MVD） and insulin-like growth factor 1 （IGF-1） were detected by immunohistochemical method.Results In comparison with sham operation group

MVC

MVD and IGF-1 increased significantly in model group （P<0.05）.MVC and MVD in large dose YHF group increased significantly compared with other groups（P<0.05） except small dose YHF group.There was significant difference in MVC between small dose YHF group and model group（P<0.05）.The expression of IGF-1 in both large and small dose YHF groups were higher than that in model group（P<0.05）.Conclusion Large dose of YHF can enhance angiogenesis and protect myocardium in ischemic myocardium of rats after myocardial infarction

which may be relevant to its up-regulating the expression of IGF-1.
