Metabonomics Study on Phlegm and Blood Stasis Evolution of Hyperlipidemia and Atherosclerosis
Objective To explore the dynamic evolution and phase characteristics of phlegm and blood stasis and the conditions of evolution from phlegm to stasis for studying its biological features from the lipid metabolism. Methods Seventy-two male Wistar rats were randomly divided into the control group and model group with 36 in each. The hyperlipidemia models were established by feeding high fat forage. At the second
4th and 8th week
plasma was collected from the abdominal aorta and detected by nuclear magnetic resonance(NMR)spectrometer for the principal component analysis(PCA).Results The PCA of rat plasma 1H NMR spectra revealed different metabolic spectra between model and control groups. With the time prolonging of model making
the different metabolic spectra were showing lipid metabolism disorder at the early stage of hyperlipidemia
later the appearance of acetyl glycoproteins which involved in the blood coagulation
and then the increase of lactate and lactic acid which meant the aggravation of lipid metabolism disorders
consistent with the changes of blood lipids and hemorheological parameters. Conclusion Characteristic metabolic products in the evolution from phlegm to stasis can be identified by NMR based metabonomics analysis to interpret the biological features of TCM syndromes.
