摘要

目的观察脑心通对氧化低密度脂蛋白（ox-LDL）作用下的人THP-1细胞活力及炎症反应的影响。方法将THP-1源性巨噬细胞分为7个组

即对照组（不加任何干预）、ox-LDL组（50mg/L ox-LDL干预细胞24h）、脑心通1组、脑心通2组、脑心通3组、脑心通4组（分别以0.125g/L、0.25g/L、0.5g/L、1.0g/L脑心通和50mg/L ox-LDL与细胞共孵育24h）、阿托伐他汀组（1μmol/L阿托伐他汀和50mg/L ox-LDL与细胞共孵育24h）。噻唑蓝（MTT）比色法检测THP-1细胞活力

酶联免疫吸附法（ELISA）检测细胞上清液中白细胞介素6（IL-6）和肿瘤坏死因子α（TNF-α）的含量。结果脑心通有轻度保护ox-LDL作用下巨噬细胞活力的作用

并抑制其引起的细胞炎症反应

上述作用均呈浓度依赖性

1g/L脑心通作用最为明显。结论脑心通呈浓度依赖性抑制ox-LDL引起的细胞损伤及炎症反应。

Abstract

Objective To investigate the effect of Naoxintong Capsule（Chinese prepared medicine for atherosclerosis） on cell vitality and inflammatory reaction of THP-1 macrophages induced by oxidized low density lipoprotein（ox-LDL）.Methods The THP-1 original macrophages were divided into seven groups: control group

ox-LDL group（50mg/L ox-LDL intervention for 24h）

Naoxintong group Ⅰ

Ⅱ

Ⅲ

Ⅳ（0.125g/L

0.25g/L

0.5 g/L

1.0 g/L Naoxintong and 50mg/L ox-LDL incubated with macrophages for 24h）

atorvastatin group（atorvastatin 1μmol/L and 50mg/L ox-LDL incubated with macrophages for 24h）.THP-1 macrophages vitality was tested with MTT colorimetric method and the contents of IL-6 and TNF-α were detected by enzyme linked immunosorbent assay respectively.Results Naoxintong has slight cytoprotection effect and inhibits inflammatory reaction of THP-1 macrophages induced by ox-LDL in a mode of dose-dependence

and the most obvious effect was in a dose of 1.0g/L.Conclusion Naoxintong slightly inhibits the cytotoxicity and inflammatory reaction of THP-1 macrophages induced by ox-LDL in a dose-dependent effect.
