摘要

目的探讨筋脉通胶囊对糖尿病周围神经病变的作用及其机制。方法以Wistar大鼠按成人剂量15倍给药分别制成筋脉通和神经妥乐平含药血清。实验分为空白对照组（不加细胞）、正常对照组（加入正常大鼠血清）、高糖组（加入葡萄糖）、筋脉通组（加入筋脉通含药血清和正常大鼠血清）、神经妥乐平组（加入神经妥乐平含药血清）

除空白对照组和正常对照组之外

其余各组均加入50mmol/L葡萄糖培养高糖雪旺细胞模型。采用免疫荧光法检测高糖培养雪旺细胞睫状体神经营养因子（CNTF）的表达

实时荧光定量PCR法检测CNTF-mRNA的表达。结果与正常对照组相比

高糖组、筋脉通组、神经妥乐平组雪旺细胞CNTF及CNTF-mRNA的表达均降低（P<0.01）;与高糖组相比

筋脉通组与神经妥乐平组雪旺细胞CNTF及CNTF-mRNA表达均增高（P<0.01）;且筋脉通组雪旺细胞CNTF-mRNA表达较神经妥乐平组增高（P<0.05）。结论筋脉通胶囊可以上调高糖培养雪旺细胞CNTF蛋白及其mRNA的表达

从而对糖尿病周围神经病变具有一定的改善作用。

Abstract

Objective To investigate the effect of Jinmaitong Capsule（Herbal capsule for removing obstruction of channels）on diabetic peripheral neuropathy and its machanism.Methods The Jinmaitong Capsule and Neurotropin medicated serum was prepared respectively according to the 15 times of adult dosage for Wistar rats.In the experiment

the grouping was the blank control group（without cells added）

normal control group（with normal rat serum added）

high glucose group（with glucose added）

Jinmaitong group（with the Jinmaitong Capsule medicated serum and normal rat serum added）

and neurotropin group（with the neurotropin medicated serum added）.Except the blank control group and normal control group

the 50 mmol/L glucose was added to all other groups respectively to make high glucose Schwann cell models.The expression of ciliary neurotrophic factor（CNTF）and CNTF-mRNA was detected by SABC immunohistochemistry method and real-time fluorogenetic quantitative PCR respectively.Results Compared with the normal control group

the CNTF and CNTF-mRNA expression in the high glucose group

Jinmaitong group

and neurotropin group was lowered（P<0.01）.Compared with the high glucose group

the CNTF-mRNA and CNTF expression of the Jinmaitong group and neurotropin group was increased（P<0.01）and the CNTF-mRNA expression in the Jinmaitong group was higher than that in the neurotropin group（P<0.05）.Conclusion Jinmaitong Capsule can up-regulate the expression of CNTF and CNTF-mRNA of rat Schwann Cells cultured in high glucose medium

so as to improve the diabetic peripheral neuropathy.
