摘要

目的观察消膏转浊方对肥胖型非酒精性脂肪肝大鼠胰岛素抵抗的影响。方法将40只Wistar大鼠随机分为5组。正常组以标准饲料喂养;其余组大鼠饲以高脂饲料。8周后

消膏转浊方组以及罗格列酮组改为普通饲料喂养

同时给予药物治疗。饮食控制组改为普通饲料喂养

共12周。于禁食次日进行钳夹实验

并计算葡萄糖输注率（GIR）。实时荧光定量PCR方法检测大鼠肝脏胰岛素受体（InsR）和胰岛素受体底物-2（IRS-2）的基因表达。结果与模型组比较

消膏转浊方组与罗格列酮组均能够显著提高肥胖型非酒精性脂肪肝大鼠的GIR（P<0.01）

其InsR mRNA、IRS-2 mRNA的表达均显著高于模型组（P<0.01）。结论消膏转浊方通过增加肝脏组织InsR以及IRS-2的表达来改善胰岛素抵抗状态。

Abstract

Objective To observe the effect of Xiaogao Zhuanzhuo Formula （Herbal formula for dissolving fat） on the insulin resistance of rats with obese non-alcoholic fatty liver. Methods Forty Wistar rats were randomized into 5 groups. The normal group was fed with standard feed and the rest were with high fat feed. Eight weeks later

the Xiaogao Zhuanzhuo Formula group and Rosiglitazone group were fed with ordinary feed and treated with the corresponding medicine. The food control group was fed with ordinary feed for 12 weeks. On the following day of fasting

clamp experiment was conducted and glucose infusion rate （GIR） was calculated. The expression of liver tissue insulin receptor （InsR） and its substrate-2 （IRS-2） was determined with the real-time fluorescence quantitative PCR detection method. Results Compared with those in the model group

GIR was increased significantly （P<0.01） and the expression of InsR mRNA and IRS-2 mRNA was significantly higher （P<0.01） in both Xiaogao Zhuanzhuo Formula group and Rosiglitazone group. Conclusion Xiaogao Zhuanzhuo Formula improves the state of insulin resistance through increasing the expression of InsR and IRS-2 of liver tissues of rats.
