摘要

目的观察补肾法、疏肝法对超促排卵小鼠卵母细胞数量及质量的影响。方法将90只雌性昆明小鼠随机分为对照组、正常组、补肾高剂量组、补肾低剂量组、疏肝高剂量组、疏肝低剂量组各15只

补肾高、低剂量组分别给予补肾调经方精制口服液5.4、2.7g/ml

疏肝高、低剂量组分别给予逍遥丸混悬液0.6、0.3g/ml

对照组和正常组用蒸馏水灌胃

各组小鼠按均1ml/100g体重灌胃

连续10d。第11日除正常组外其余各组小鼠腹腔注射孕马血清促性腺激素

48h后腹腔注射人绒毛膜促性腺激素

16h后测定各组小鼠血清中雌二醇（E2）、促黄体生成激素（LH）和促卵泡生成激素（FSH）的水平

比较各组排卵数、优质卵泡数

检测卵母细胞中生长分化因子-9（GDF-9）蛋白和GDF-9mRNA的表达。结果各给药组FSH、LH、E2表达和GDF-9蛋白及其mRNA显著高于对照组（P<0.05）;并且补肾高剂量组和疏肝高剂量组均显著高于补肾低剂量组和疏肝低剂量组（P<0.05）。各给药组优质卵泡数均高于对照组（P<0.05）。对照组及各给药组排卵数较正常组显著增加（P<0.05）。结论补肾法、疏肝法可增加小鼠卵母细胞数量、提高优质卵泡率、促进卵子排出

其作用机制可能与调控卵母细胞GDF-9表达相关。

Abstract

Objective To observe the effects of kidney-supplementing therapy and liver-coursing therapy on the quantity and quality of oocytes in superovulated mice.Methods Ninety female Kunmin mice were randomized into control group

normal group

kidney-supplementing high-dose group

kidney-supplementing low-dose group

liver-coursing high-dose group and livercoursing low-dose group

with 15in each.The kidney-supplementing high-dose group and kidney-supplementing low-dose group was given 5.4g/ml and 2.7g/ml refined oral solution of kidney-supplementing menstruation-regulating formula respectively.The liver-coursing high-dose group and liver-coursing low-dose group was given 0.6g/ml and 0.3g/ml suspension of Xiaoyao Pill（Free Wanderer Pill）.The control group and normal group were given distilled water intragastrically.The mice in each group were gavaged with 1ml/100g for 10consecutive days.On the 11th day

mice in groups except the normal group were peritoneally injected gonadotropin from pregnant horse

and 48hours afterwards

human chorionic gonadotropin was injected peritoneally.The serum estradiol（E2）

luteinizing hormone（LH）and follicle-stimulating hormone（FSH）was detected after 16hours.The ovulation rates and quality follicle counts were compared.The expressions of growth and differentiation factor-9（GDF9）protein and GDF9mRNA in oocytes were detected.Results Comparing with the control group

the expressions of FSH

GDF9and GDF9mRNA were significantly increased in other groups（P<0.05）

especially in two high-dose administration groups as comparing with two low-dose administration groups（P<0.05）.The quality follicle counts in the administration groups were significantly higher than that in the control group（P<0.05）.Comparing with the normal group

the ovulation rates in the control group and administration groups were significantly increased（P<0.05）.Conclusion The kidney-supplementing therapy and liver-coursing therapy can increase the quantity of oocytes

improve the rate of quality follicular and promote ovum

which may be relevant to regulating the expression of GDF9in oocytes.
