In Vitro and In Vivo Anti-Tumor Effects of Ganoderma Compound
Objective To observe the anti-tumor effects of Ganoderma Compound and its possible mechanism. Methods The in vitro cell culture method was used to observe the inhibitory effect of Ganoderma Compound on the proliferation of human hepatoma cell line QGY7703
rat hepatoma cell line CBRH7919 and human gastric cancer cell line BGC823. In vivo experiment fifty mice were randomized into the normal control group
model group
Cyclophosphamide group
Ganoderma Compound group and combo therapy group
with 10 in each. The S180 tumor-bearing mouse models were established except the normal control group. The Cyclophosphamide group was given intraperitoneal injection of 30 mg/kg Cyclophosphamide. The Ganoderma Compound group was gavaged with 50 mg/kg Ganoderma Compound. The combo therapy group was given Cyclophosphamide and Ganoderma Compound. The normal control group and model group were given the same amount of normal saline. Each group was administered for 14 continuous days. The tumor inhibition rate
spleen index and thymus index was calculated. The serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected. Results The drug concentration of 0.4 to 6.4 mg/ml in the Ganoderma Compound group had a significantly inhibitory effect on the proliferation of CBRH7919 and BGC823 tumor cell lines with a concentration dependent manner (P<0.05). The tumor inhibition rate in the Ganoderma Compound group
Cyclophosphamide group and combo therapy group was 47.90%
69.57% and 78.67% respectively. Comparing with the Cyclophosphamide group
the spleen index and thymus index in the Ganoderma Compound group and combo therapy group was significantly increased (P<0.05). The levels of TNF-α and IL-6 in three administration groups were significantly higher than those in the normal control group except for the IL-6 level in the Cyclophosphamide group (P<0.05). Conclusion The Ganoderma Compound has a certain anti-tumor activity and its mechanism may be related to enhancing the immunity and improving the serum contents of TNF-α and IL-6 in tumor-bearing mice.
