Effect of Sequential Yin-Nourishing Yang-Supplementing Medicinal on Ovarian Gene Expression Profiles in Light Damaged Rats
Objective To study the effect and mechanism of sequential yin-nourishing yang-supplementing medicinal on ovarian differential gene expression in rats with reproductive rhythm disorders under artificial light environment. Methods The rat models of reproductive rhythm disorders were established under artificial light environment.Thirty rats were randomized into the blank group
model group and Chinese medicinal group
with 10in each. The Chinese medicinal group was sequentially administered with 1ml/100g yin-nourishing formula and yang-supplementing formula. The model group and blank group were gavaged with same volume of normal saline. The intragastric administration lasted for 18days in each group. The total RNA in ovarian tissue of rats was extracted with one-step method and fluorescence labeled with Cy3and Cy5after reverse transcription. The ovarian cDNA probes of rats in each group were got and then hybridized with cDNA microarray. The results were scanned with laser scanner and the Ratio analysis and cluster analysis was conducted with software. Results Cluster analysis showed that the genetic profile in the Chinese medicinal group and model group was quite different. There were 1 896 differentially expressed genes(1 065down-regulating genes and 831up-regulating genes)as the model group compared with normal group. There were 2 582differentially expressed genes(1 325down-regulating genes and 1 257up-regulating genes)as the Chinese medicinal group compared with model group. The deviation of 1 067differentially expressed genes had been adjusted with increased expression in 611genes and decreased expression in 456genes.Differentially expressed genes in biological processes and pathway analysis showed that the regulation of Chinese medicinal on the genetic network expression of luteinizing hormone(LH)receptor and melatonin(MT)receptor was obvious. Conclusion The sequential yin-nourishing yang-supplementing medicinal can partially alleviate light-induced reproductive rhythm disorders and improve ovarian function by regulating cell proliferation and inhibiting apoptosis and the anabolism of steroids.
