摘要

目的探讨益气养阴方治疗急性髓细胞白血病的可能作用机制。方法 40只NOD-SCID小鼠经60Co照射后

尾静脉注射KG1a细胞1×106个

建立急性髓细胞白血病动物模型后随机分成4组

每组10只。模型组予生理盐水灌胃;扶正组予扶正方浓缩药液（1.08g/ml）灌胃;祛邪组予祛邪方浓缩药液（0.92g/ml）灌胃;全方组予益气养阴方浓缩药液（2g/ml）灌胃。各组均0.2ml/次

每日2次

共28天。采用PCR-ELISA法检测骨髓细胞端粒酶活性;荧光定量PCR法检测人端粒酶逆转录酶（hTERT）-mRNA及其启动子甲基化的表达。结果扶正组、祛邪组和全方组小鼠骨髓细胞端粒酶活性、hTERT-mRNA和hTERT启动子甲基化的表达较模型组明显降低（P<0.05或P<0.01）

全方组端粒酶活性、hTERT-mRNA和hTERT启动子甲基化的表达低于扶正组与祛邪组（P<0.05或P<0.01）。结论益气养阴方治疗急性髓细胞白血病的机制可能与降低端粒酶活性、hTERT及其启动子甲基化的表达水平有关。

Abstract

To research the possible mechanism of Yiqi Yangyin Formula（ Qi-Boosting Yin- Nourishing Formula） for acute myeloid leukemia（ AML）. Methods Forty nonobese diabetic / severe combined immunodeficient（ NOD-SCID） mice were irradiated with60Co and induced into AML models with tail vein injection of 1 × 106KG1a cells. The models were randomized into four groups with 10 in each. The model group was gavaged with normal saline. The vital qi-supporting group was gavaged with 1. 08 g / ml concentrated solution of vital qi-supporting formula. The evil-dispelling group was gavaged with 0. 92 g / ml concentrated solution of evil-dispelling formula. The entire formula group was gavaged with 2 g / ml concentrated solution of entire formula. Each group was gavaged with 0. 2 ml solution / each time

twice daily for 28 days. The telomerase activity in bone marrow cells was detected with PCR-ELISA method. The methylation and expression of human telomerase reverse transcriptase（ hTERT） mRNA and its promoter were detected with quantitative PCR. Results Comparing with the model group

the activity of hTERT and the methylation and expression of hTERT mRNA and its promoter was significantly decreased in the vital qi- supporting group

evil-dispelling group and entire formula group（ P < 0. 05 or P < 0. 01）

especially in the entire formula group（ P < 0. 05 or P < 0. 01）. Conclusion Yiqi Yangyin Formula can treat AML by reducing the activity of hTERT and the methylation and expression of hTERT mRNA and its promoter.
