Effects of Qingre Ditan Jiejing Fang( 清热涤痰解痉方) on Serum ox-LDL,hs-CRP and ET-1 of Coronary Artery Spasm Mice
Objective To observe effects of Qingre Ditan Jiejing Fang( 清热涤痰解痉方) on mice with hyperlipemia and coronary artery spasm induced by pituitrin and study the possible mechanism. Methods Totally 140C57 BL /6J mice were randomly divided into normal group
model group
diltiazem group
atorvastatin group and Chinese medicine high
medium and low dose group
with 20 mice in each group. Except for the normal group
the rest groups were given high-fat fodder to feed for 4 weeks. The Chinese medicine high
medium and low dose group were respectively given granules of Qingre Ditan Jiejing Fang
7. 2
3. 6
1. 8 g /( kg·d). The diltiazem group was given diltiazem
0. 04 g /( kg·d). The atorvastatin group was given atorvastatin calcium tablet
0. 013 g /( kg · d). The normal group and the model group were given 0. 5% sodium carboxymethyl cellulose solution with same volume
once a day for four weeks. One hour after the last given treatment
the mice were given intraperitoneal injection with pituitrin 60 U / kg to establish model of coronary artery spasm. The researchers recorded Ⅱ-lead electrocardiogram at the time that 10 min before and 5
10
15 min after injection and counted displacement of T wave. The researchers detected level of serum oxidized low density lipoprotein( ox-LDL)
high-sensitivity C-reactive protein( hs-CRP) and endothelin-1( ET-1) after test. Results At the time that 5
10
15 min after intraperitoneal injection with pituitrin
in the model group
displacement of T wave in electrocardiogram significantly increased compared with the normal group( P < 0. 05); Compared with the model group
the diltiazem group
the atorvastatin group and the Chinese medicine high
medium and low dose group at above three time all can lower displacement of T wave( P < 0. 05); Compared with the atorvastatin group
displacement of T wave significantly decreased in the Chinese medicine high and medium dose group at the 5
10 min and the Chinese medicine high dose group at the 15 min( P < 0. 05). Compared with the normal group
serum ox-LDL
hs-CRP
ET-1 in the rest groups significantly increased( P < 0. 05 or P < 0. 01).Compared with the model group
ox-LDL in the Chinese medicine high dose group and the atorvastatin group significantly decreased
hs-CRP in each group that been given medicines significantly decreased and ET-1 in the diltiazem group and the Chinese medicine high dose group significantly decreased( P < 0. 05). Conclusions Qingre Ditan Jiejing Fang may effectively relieve pituitrin inducing coronary artery spasm of mice with hyperlipemia and its mechanism may be related to lowering serum ox-LDL
