Effect of Huiyang Shengji Ointment(回阳生肌膏) Components on Skin Wound Healing and Macrophage Phenotype Transformation in Chronic Skin Ulcer Model Mice
Objective To observe the effect and possible functional mechanism of Huiyang Shengji Ointment on chronic skin ulcer. Methods Divided 150 BALB / c mice into blank group
model group
western medicine group
Tonifying Qi and Warming Yang group( TQWY)
Activating Blood and Dredging Collaterals group( ABDC)
with30 mice in each group. Adopted immunosuppression combined with skin wound method to build mice of chronic skin ulcer model. The TQWY group and the ABDC group were respectively given Yiqi Wenyang Formula and Huoxue Tongluo Formula 0. 018 g each
while the western medicine group was given recombinant human basic fibroblast growth factor 0. 3 g
which were all made into medicated gelatin sponge applied with sterile patch. Changed medicine every other day and took photos of skin wound. Measured square of skin wound and calculated healing rate; took skin wound tissue at the 3rd
7th and 14 th of wound and observed new granulation tissue
collagen distribution and so on in skin wound using HE
Masson and sirius red staining; examined number of macrophagocyte in skin wound tissue using immunohistochemical method; examined the expression of genes of cytokines secreted by M1 and M2 type macrophagocyte in local skin wound using real time PCR. Results Compared with the model group
skin wound areas in the TQWY group and the ABDC group decreased and healing rate increased on the 7th and the 14 th day of injury( P < 0. 05). And healing rate in the ABDC group was higher than that in the TQWY group( P < 0. 05). On the 3rd day of wound
interleukin 6( IL-6) levels in the blank group
western medicine group and TQWY group was lower than that in the model group
while vascular endothelial growth factor( VEGF) and interleukin 10( IL-10) level were higher than that in the model group. VEGF and IL-10 level in the ABDC group was higher than that in the model group. VEGF level in the ABDC group was higher than that in the TQWY group( P < 0. 05). On the 7th day of wound
M1 type macrophage number of skin and TNF-αlevels in the blank group
western medicine group
the TQWY group and the ABDC group were remarkably lower than that in the model group. M2 type macrophage number
VEGF and IL-10 level was higher than the model group( P < 0. 05 or P < 0. 01). VEGF and IL-10 level in the ABDC group was higher than that in the TQWY group( P < 0. 05). Conclusion Huoxue Tongluo Formula and Yiqi Wenyang Formula could both promote chronic skin wound healing in mice. Its mechanism may be promoting wound healing transferring from inflammation phase to proliferating phase by promoting macrophage phenotypic transformation. Huoxue Tongluo Formula had better effect in macrophage phenotypic transformation than Yiqi Wenyang Formula.
