Relations between Macroscopic Characterization and Metabolomics of Rat with Hypertension of Ascendant Hyperactivity of Liver Yang Syndrome Intervened with Gouteng(Uncariae Ramulus Cum Uncis,钩藤)
Objective To explore the possible mechanism of Gouteng(Uncariae Ramulus Cum Uncis
钩藤) in treating hypertension of ascendant hyperactivity of liver Yang syndrome. Methods Fourteen SHR rats were randomized into ascendant hyperactivity of liver Yang syndrome(AHLY) group and Gouteng group
with 7 rats in each group. Another seven Wistar rats were selected as control group. AHLY group and Gouteng group were given oral Fuzi(Aconiti Lateralis Radix Praeparata
附子) solution of 20 ml/kg to build the hypertension rat model of AHLY syndrome. After successful modeling
Gouteng group was given 22. 9 ml/kg Gouteng decoction
while the AHLY group and the control group were respectively given the same amount of normal saline; the rats in each group were intervened for 4 weeks. Then the macro-indicators such as systolic blood pressure
facial temperature
and pain threshold and rotation time were collected. At the same time
the information of endogenous metabolites in serum of rats was collected by UHPLC-quadrupole-electrostatic field orbitrap high resolution mass(UPLC-Q-Exactive) technology. Partial least-squares regression was used to analyze the relationship between external macro-indicators and internal microindicators. Results Compared with those in the control group
the systolic blood pressure increased
the facial temperature increased
the pain threshold decreased and the rotation time shortened in the model group
while the systolic blood pressure in Guoteng group was lower than that in the AHLY group
and the rotation time was longer(P < 0. 05 or P < 0. 01). A total of 16 biomarkers were identified by database search
including linoleic acid
dopamine
deoxycorticosterone
4-hydroxyproline
dihydrotestosterone
11 b-hydroxyprogesterone
glycylglycine
6-ketone prostaglandin
16 a-hydroxyandrostone
L-tryptophan
leukotriene B4
thromboxane B2
phosphatidic acid
2-methoxy estrone
uric acid
and lysophosphatidylcholine. Compared with those in the control group
the levels of dihydrotestosterone
11 b-hydroxyprogesterone
deoxycorticosterone
2-methoxy estrone decreased(P < 0. 05 or P <0. 01)
and the rest of the biomarkers increased in the AHLY group(P < 0. 05 or P < 0. 01); compared with those in the AHLY group
the levels of dihydrotestosterone
16 a-hydroxyandrostaglandin
deoxycorticosterone and 2-methoxy estrone in Gouteng group increased(P < 0. 05 or P < 0. 01)
and the rest of the biomarkers decreased(P < 0. 05 or P < 0. 01). Conclusion The possible mechanism of Gouteng in treating hypertension of the ascendant hyperactivity of liver Yang syndrome may include the following: regulating the metabolisms of glycine
serine and threonine
regulating the metabolisms of arachidonic acid and tyrosine to reduce facial temperature
regulating the metabolisms of glycerophospholipid and linoleic acid and the anabolism of sterol hormones to improve dizziness as well as regulating metabolisms of arachidonic acid
tyrosine and purine to improve the pain threshold.
