Impact of Needling Point “Renying(ST9)” on Hypothalamic Gene Expression of Spontaneous Hypertension Rats
Objective To explore the possible mechanism of needling Point Renying( ST 9) in treating hypertension. Methods Twenty spontaneous hypertension rats( SHR) were randomized into a Renying group and a model group( n = 10 in each group). Another ten Wistar-Kyoto( WKY) rats were set as a blank group. The rats in Renying group were needled at Point "Renying( ST9) "for 20 min each day
taking one day off every 6 days
totally for 28 days. The model group and the blank group were given no intervention. Rats’ blood pressure was measured on the 1 st
5 th
9 th
13 th
17 th
21 st
25 th and 28 th day of needling. At the end of the experiment
hypothalami was collected to make gene chip analysis. The expressions of key gene 5-hydroxytryptamine 1 A receptor( 5-HT1 A) and Cystathionine β synthetase( CBS) mRNA were detected with polymerase chain reaction( PCR) technology. Results Compared with that in the blank group
systolic pressure of rats in the model group increased on the 1 st
5 th
9 th
13 th
17 th
21 st
25 th and 28 th day( P < 0. 01). Compared with that in the model group
systolic pressure of rats in the Renying group decreased on the 5 th
9 th
13 th
21 st
25 th and 28 th day( P < 0. 05 or P < 0. 01). Compared with that at the previous time point in the same group
blood pressure in the model group increased on the 21 st
25 th and 28 th day
while blood pressure in the Renying group increased only on the 21 st day( P < 0. 05). Compared with those in the blank group
129 genes were up-regulated
and 243 genes were down-regulated in the model group( P <0. 05). Compared with those in the model group
167 genes were up-regulated
and 188 genes were down-regulated in the Renying group( P < 0. 05). Compared with those in the blank group
the expressions of 5-HT1 A mRNA and CBS mRNA in the model group decreased( P < 0. 01). Compared with those in the model group
the expressions of5-HT1 A mRNA and CBS mRNA in the Renying group increased( P < 0. 01). Conclusion Needling Point"Renying( ST9) "seems to be effective in decreasing blood pressure of SHR. Influencing hypothalamus gene expression
