Protective Effects of Qingxuan Jiangya Decoction (清眩降压汤) on Kidney Damage in Spontaneously Hypertensive Rats
Objective To explore the protective effects and mechanism of Qingxuan Jiangya Decoction( 清眩降压汤) on kidney damage which resulted from essential hypertension. Methods A total of 20 spontaneously hypertensive rats( SHR) were randomly divided into model group and Qingxuan Jiangya Decoction( QXJYD) group
with10 rats in each group
and other 10 Wistar rats were taken as normal group. The rats in QXJYD group were treated with 5. 4 g/( kg·d) Qingxuan Jiangya Decoction intragastric administration while the model group was given 1 ml/d normal saline( NS)
and no treatment was performed in normal group. Systolic blood pressure of all groups rats was detected weekly with noninvasive blood pressure meter. After 8 weeks
the pathological changes of kidney tissue
nicotinamide oxidase 2( NOX2) and nicotinamide oxidase 4( NOX4) in oxidative stress level in renal tissue and the expression of interleukin 6( IL-6)
tumor necrosis factor α( TNF-α) were observed. Results During the 1-8 week of treatment
the systolic blood pressure of the model group was significantly higher than that of the normal group( P < 0. 05). Since the 3 rd week of treatment
the systolic blood pressure in QXJYD group was significantly decreased compared with the model group( P < 0. 05). The renal tubular injury in QXJYD group was improved more significantly than that in the model group. Compared with the normal group
the expressions of NOX2
NOX4
IL-6
TNF-αwere decreased more in the model group( P < 0. 05)
and above expression levels in QXJYD group were decreased more than those in model group( P < 0. 05). Conclusion Qingxuan Jiangya Decoction is effective in decreasing SHR systolic pressure and protecting the renal tubular tissues from injury. The mechanism might be related to reducing oxidative stress and inflammatory level.
