摘要

目的探讨补肾活血方治疗慢性心力衰竭的可能作用机制。方法 60只SD大鼠随机选出15只为假手术组

其余45只采用冠状动脉结扎及术后给予减食和力竭式游泳建立慢性心力衰竭大鼠模型

造模成功后随机分为模型组、西药组、中药组。假手术组和模型组大鼠每日给予蒸馏水灌胃2次

每次3 ml;西药组给予卡托普利片12. 5 mg/（kg·d）灌胃;中药组给予补肾活血方9. 2 g/（kg·d）灌胃

持续给药4周。实验期间观察各组大鼠皮毛、活动、体重、喘鸣音等一般情况

心脏彩色超声检测大鼠舒张末期左心室内径（LVEDD）、收缩末期左心室内径（LVESD）

并计算左心室射血分数（EF）及左心室短轴缩短分数（FS）

测定大鼠血清脑利钠肽（BNP）水平和大鼠肾脏组织中水通道蛋白2 （AQP2）表达。结果模型组大鼠毛色晦暗

甚至脱毛

活动性差

体重减轻

部分大鼠有咳嗽、喘促症状;而西药组、中药组大鼠各症状均有所改善。与假手术组比较

模型组大鼠LVEDD、LVESD升高

EF、FS降低

血清BNP水平和肾组织AQP2表达升高（P <0. 05）。与模型组比较

中药组及西药组大鼠LVEDD、LVESD降低

EF、FS升高

血清BNP水平及肾组织AQP2表达降低（P <0. 05）;西药组与中药组各指标比较差异无统计学意义（P> 0. 05）。结论补肾活血方可能通过降低慢性心力衰竭大鼠血清BNP

下调肾组织AQP2表达

从而改善心功能。

Abstract

Objective To investigate the possible mechanism of Bushen Huoxue Recipe（补肾活血方） in the treatment of chronic heart failure. Methods A total of 15 Sprague-Dawley rats were randomly selected from 60 rats into sham operation group. The remaining 45 rats underwent coronary artery ligation

post-operative food reduction and exhaustive swimming to establish a rat model of chronic heart failure. After successful modeling

they were randomly divided into model group

western medicine group

and Chinese medicine group. Rats in the sham operation group and the model group were given 3 ml of distilled water

twice a day; the western medicine group was given12. 5 mg/（kg·d） of captopril tablets by gavage; the Chinese medicine group was given 9. 2 g/（kg·d） of Bushen Huoxue Recipe by gavage for 4 weeks. During the experiment

the general condition of fur

activity

body weight and wheezing sound of each group were observed. Color Doppler echocardiography was used to detect left ventricular enddiastolic dimension（LVEDD）

left ventricular end-systolic dimension（LVESD）

left ventricular ejection fraction（EF） and left ventricular short-axis shortening fraction（FS） were calculated. The serum brain natriuretic peptide（BNP） levels and aquaporin 2（AQP2） expressions in rat’s kidney tissue were determined. Results In the model group

the fur color was dark with hair loss. The activity was poor

and body weight was reduced. Some rats had symptoms of cough and wheeze. However

the symptoms of rats in the western medicine group and the Chinese medicine group were improved. Compared with the sham operation group

LVEDD and LVESD were increased

EF and FS decreased

and serum BNP level and AQP2 expression in the kidney tissue increased in the model group（P <0. 05）. Compared with the model group

LVEDD and LVESD were decreased

EF and FS increased

and serum BNP level and AQP2 expression in the kidney tissue were decreased in the Chinese medicine group and western medicine group（P < 0. 05）. There was no significant difference between the western medicine group and Chinese medicine group（P > 0. 05）. Conclusion Bushen Huoxue Recipe may improve cardiac function by reducing serum BNP in rats with chronic heart failure and down-regulating the expression of AQP2 in renal tissue.
