Evaluation on Sleep Deprivation Combined with Left Anterior Descending Coronary Artery Ligation to Establish Rat Model of Heart Failure with Qi Deficiency and Blood Stasis Syndrome
Objective To evaluate sleep deprivation combined with left anterior descending coronary artery ligation to establish rat model of heart failure with qi deficiency and blood stasis syndrome. Methods A total of 70 SD rats were randomly divided into a blank control group
a model group
a sleep deprivation group
a myocardial infarction group
a Yiqi Huoxue Granule(益气活血颗粒) groups with high
medium and low dosage
with 10 rats in each group. The sleep deprivation group was given irregular sleep deprivation for 8 weeks. The rats in the model group and Yiqi Huoxue Granule group were given 3 weeks of sleep deprivation
and the left anterior descending coronary artery was ligated. The sleep deprivation continued on the 2 nd day after surgery. A model of qi deficiency and blood stasis syndrome of heart failure was established at 5 weeks. Only the left anterior descending coronary artery was ligated in the myocardial infarction group
and the blank control group was fed normally. On the 2 nd day after the operation
Yiqi Huoxue Granules were given gavage at 4.0
2.0
and 1.0 g/(kg·d) in the high
medium
and low-dose groups
and the other groups were given saline 1 ml/100 g
for 5 weeks. The body weight
grasping force
pulse amplitude
autonomic activity
tongue image
cardiac function
ventricular remodeling index
hemorheology index and abdominal aortic blood flow velocity of the rats in each group were compared. Results Compared with the blank control group
the body weight
grasping force
pulse amplitude
and total distance and average speed of autonomous activities of the model group significantly decreased
and the index of ventricular remodeling increased
and the index of cardiac function
wall thickness
and abdominal aortic blood flow velocity significantly decreased. The blood viscosity and plasma viscosity increased significantly under different shear rates
and the relative values of R(red)
G(green)
and B(blue) on the tongue surface were significantly reduced(P<0.05 or P<0.01). Compared with the model group
the Yiqi Huoxue Granule high-dose group can increase rat weight
grasping force
pulse amplitude
wall thickness
reduce ventricular remodeling
hemorheology indexes
improve rat heart function and autonomous behavior activities
and increase abdominal aorta blood flow velocity
and the relative values of R and G on the tongue surface of rats significantly increased(P<0.05 or P<0.01). Conclusion The combination of sleep deprivation and left anterior descending coronary artery ligation can induce heart failure rats to show obvious external and objective manifestations of qi deficiency and blood stasis syndrome. The effective intervention of high-dose Yiqi Huoxue Granules contradicts the scientificity and reliability of this model.
