Effect of Xiaopi Granules (消痞颗粒) on miR-21-PTEN-PI3K/AKT Signaling Pathway in Gastric Mucosa Tissue of Precancerous Lesions of Gastric Cancer Model Rats
Objective To explore the possible mechanism of Xiaopi Granules(消痞颗粒) in treating precancerous lesions of gastric cancer(PLGC). Method A total of 70 Wistar rats were randomly divided into a blank group of 10 rats
and 60 rats for making models. The modeling group is made by gavage with 200μg/ml of N-methyl-N′-nitro-N-nitrosoguanidine solution 5 ml/(kg·d) each
free drinking of 0.05% ammonia solution and pellet feed containing 0.03% Ranitidine. The 3 factors were combined to establish a rat model of PLGC. At the end of the 44 th week
41 successful modeling rats were randomly divided into a model group(14 rats)
a vitacoenayme group(14 rats)
and a Xiaopi Granules group(13 rats). Rats in the model group were given normal saline 3 ml/(kg·d) by gavage
the vitacoenayme group was gavage with vitacoenayme tablets 0.3 g/(kg·d)
and the Xiaopi Granules group was gavage with Xiaopi Granules 9 g/(kg·d) for 12 weeks. RT-PCR was used to detect the expression of miR-21 and PTEN gene in gastric mucosa of rats
and phosphoinositide 3-kinase(PI3 K)
protein kinase B(AKT) protein expression in gastric mucosa of rats was detected by ELISA. Results The differences in the expression of miR-21
PTEN gene and AKT
PI3 K protein in gastric mucosal tissue of rats in each group were statistically significant(P<0.01). Compared with the blank group
the expression of miR-21 gene in the model group and the vitacoenayme group increased
and the expression of PTEN gene decreased(P<0.01); the expression of AKT and PI3 K protein in the model group
the vitacoenayme group and the Xiaopi Granules group all increased(P<0.01). Compared with the model group
the expression of miR-21 gene in the Xiaopi Granules group was decreased
the expression of PTEN gene was increased
and the protein expression of AKT and PI3 K were decreased(P<0.01)
but there was no statistical difference in all the indicators between the model group and vitacoenayme group(P>0.05). Conclusion Xiaopi Granules may regulate the PTEN gene by inhibiting the abnormal expression of miR-21 in gastric mucosal tissue
thereby intervening the PI3 K/AKT signaling pathway to reverse the occurrence of precancerous lesions of gastric cancer.
