Effect of Electroacupuncture at “Baihui”(DU20) and “Renzhong”(DU 26) on Inflammatory Factors in Cerebrospinal Fluid and Gasdermin D Expression in Cerebral Tissue in Cortex Ischemia Area in Cerebral Ischemia Reperfusion Injury Model Rats
To explore the effect of electroacupuncture (EA) at Baihui (DU 20) and Renzhong (DU 26) on cerebral ischemia reperfusion injury (CIRI) and the possible mechanism.
Methods
2
A total of 36 male SD rats were randomized into sham-operation group (n=12), model group (n=12) and EA group (n=12). Suture middle cerebral artery occlusion was used to develop the CIRI model in all groups but the sham-operation group. Twenty-four hours after modeling, the rats in the EA group were performed for EA at Renzhong and Baihui, acupoints with a continuous wave with intensity of 1 mA and frequency of 15 Hz, once daily, 20 min each time, for 5 days. The Longa method was used to evaluate the degree of neurological function injury in the rats; TTC staining was used to measure the cerebral infarct volume of rats; HE staining was used to observe the pathological and morphological changes in brain tissue; ELISA method was used to detect the levels of interleukin 1β (IL-1β), interleukin 6 (IL-6) and interleukin 18 (IL-18) in cerebrospinal fluid; reverse transcription polymerase chain reaction and Western blotting were performed to determine the expressions of Gasdermin D (GSDMD) mRNA and the N-terminal fragment of GSDMD (GSDMD-N) protein in brain tissue.
Results
2
Compared to the sham-operation group, the model group had higher neurological function score and cerebral infarct volume, increased levels of IL-1β, IL-6 and IL-18 in the cerebrospinal fluid, as well as increased expressions of GSDMD mRNA and GSDMD-N protein (
P
<0.05 or
P
<0.01). HE staining showed that the neuron arrangement in the brain tissue of the model group was disordered; some of the nerve cells disappeared; and the nucleus was constricted with incomplete structure. Compared to the model group, the EA group had significantly decreased neurological function score, cerebral infarct volume, levels of IL-6 and IL-18 in the cerebrospinal fluid, and expression levels of GSDMD mRNA and GSDMD-N protein (
P
<0.05 or
P
<0.01); and the neurons degeneration in the cerebral cortex and the loss of the number of nerve cells were reduced.
Conclusion
2
EA at Baihui and Renzhong can reduce the cerebral infarct volume and improve the neurological function in CIRI model rats, which is likely related to the down-regulation of cerebrospinal fluid inflammatory factors and expression of GSDMD in cortical ischemic brain tissue.
