Effect of Different Stimulation Strength of Acupuncture on Pain and the Content of Prostaglandin and Oxytocin in Dysmenorrhea-like Model Rats
To observe the analgesic effect of acupuncture with different stimulation strength on dysmenorrhea-like rats and to explore the possible mechanism.
Methods
2
Forty SD female rats were randomly divided into blank group, model group, weak-stimulation acupuncture (WA) group and strong-stimulation acupuncture (SA) group, with 10 rats in each group. Dysmenorrhea-like model was established by injection of estradiol benzoate and oxytoxin in all groups except for the blank group. Rats in the blank group and the model group were only grasped and fixed without interventions; a 20-minute deep needling at bilateral sanyinjiao (SP6) acupoints with thick needles added by manipulation techniques was given to the SA group, while a 20-minute superficial needling at the same acupoints with a thin needles without manipulation techniques was given to the WA group. Writhing response of the rats was observed after withdrawal of needles. The content of prostaglandin F
2α
(PGF
2α
), prostaglandin E
2
(PGE
2
)
and oxytocin (OT) in serum and uterine tissue were detected by ELISA method, while the content of Ca
2+
in serum was detected by chemical methods.
Results
2
Compared to those in the blank group, the latent period of writhing in the model group was significantly shortened, and the writhing score was increased; the content of PGF
2a
, PGF
2a
/PGE
2
, and OT in serum and uterine tissue, and the content of Ca
2+
in serum were significantly increased; the PGE
2
content in serum and uterine tissue were reduced in the model group (
P
<0.01). Compared to the model group, the SA and WA groups had a significantly prolonged latent period of writhing, decreased writhing scores, PGF
2a
content in uterine tissue, PGF
2a
/PGE
2
and OT content in serum and uterine tissue, as well as increased PGE
2
content in serum (
P
<0.05 or
P
<0.01). The SA group had significantly decreased PGF
2a
and Ca
2+
content in serum (
P
<0.05). The latent period of writhing in the SA group was significantly longer than that in the WA group, while the writhing score was lower (
P
<0.05 or
P
<0.01), and the PGF
2a
content in serum was obviously lower in the SA group than that in the WA group (
P
<0.05).
Conclusion
2
Acupuncture could relieve the pain in dysmenorrhea-like rats, and strong-stimulation has better effects than weak-stimulation. The possible mechanism may be explained by that acupuncture can regulate the content of PGF
2a
, PGE
2
and OT in serum and uterine tissue, and reduce the content of Ca
2+
in serum, thereby improving the contraction of smooth muscle.
