Effects of Naoluoxintong and its Disassembled Prescriptions on Learning and Memory Ability and Synaptic Plasticity in Right Middle Cerebral Artery Occlusion-reperfusion Rats
To explore the possible mechanism of Naoluoxintong (脑络欣通) for prevention and treatment of ischemic stroke.
Methods
2
Fifty SD rats were randomly divided into sham operation group, model group,
Naoluoxintong
group,
Yiqi
(益气) group and
Huoxue Tongluo
(活血通络) group, with 10 rats in each group. Except for the sham operation group, the modified suture method was used in the other groups to establish the right middle cerebral ischemia-reperfusion model. After successful modeling, rats in the
Naoluoxintong
group,
Yiqi
group and
Huoxue Tongluo
group were given
Naoluoxintong
8.54 g/(kg·d),
Yiqi
Formula
3.5 g(kg·d) and
Huoxue Tongluo Formula
5.04 g/(kg·d) by gavage respectively, while the sham operation group and the model group were given the 10 ml/(kg·d) of normal saline by gavage, twice a day, for a total of 14 days. Morris water maze (positioning navigation test, spatial navigation test) was used to observe the learning and memory ability of rats, while HE staining was used to observe the pathological changes in the right frontoparietal cortex and hippocampus, and immunohistochemistry was used to detect changes in the expression of glutamate receptor 1 (GluR1) and calcium/calmodulin-dependent protein kinase Ⅱ (CaMKⅡ) in the right frontoparietal cortex and hippocampus.
Results
2
Compared with those in the sham operation group, the escape latency of rats in the model group was significantly prolonged at each time point, while the time spent in the target quadrant and the times of crossing the platform decreased, and the expressions of GluR1 and CaMKⅡ were reduced (
P
<0.01). Compared with those in the model group, the escape latency of the rats in the
Naoluoxintong
group, the
Yiqi
group and the
Huoxue Tongluo
group at each time point was significantly shortened (
P
<0.05), while the time spent in the target quadrant and the times of crossing the platform increased significantly (
P
<0.05), and the expressions of GluR1 and CaMKII increased as well (
P
<0.01). Compared with those in the
Naoluoxintong
group, the escape latency in the
Yiqi
group and the
Huoxue Tongluo
group was prolonged at each time point, while the time spent in the target quadrant and the times of crossing the platform decreased, and the expression of GluR1 and CaMKⅡ were reduced (
P
<0.05). There were no significant differences in the expression of all indicators between the
Yiq
i group and the
Huoxue Tongluo
group (
P
>0.05). The histopathological results showed that compared with those in the model group, the arrangement, shape and size of nerve cells in the
Naoluoxintong
group, the
Yiqi
group and the
Huoxue Tongluo
group were restored, and the cell edema and necrosis were improved, with more significant improvement in the
Naoluoxintong
group.
Conclusion
2
Naoluoxintong
can improve the ability of learning and memory in right middle cerebral artery occlusion-reperfusion rats, and the overall effect is better than its disassembled prescriptions. The mechanism may be related to the promotion of the expression of GluR1 and CaMKⅡ proteins in the fronto-parietal cortex and hippocampus, thereby enhancing synaptic plasticity.
