Mechanism of Action of Shenyu Yangxin Capsules (参萸养心胶囊) in Treatment of Heart Failure:Based on Transcriptomics and Experimental Validation
(参萸养心胶囊) for heart failure based on transcriptomics and experimental validation.
Method
2
Fifty C57BL/6 mice were randomly divided into a normal group (0.9% sodium chloride solution treatment), a model group (isoproterenol treatment), a low-dose
Shenyu Yangxin
group (1.56 g/kg
Shenyu Yangxin Capsules
treatment), a high-dose
Shenyu Yangxin
group (6.24 g/kg
Shenyu Yangxin Capsules
treatment), and a sodium fosinopril group (2.6 mg/kg sodium fosinopril treatment), with 10 mice in each group. All groups except the normal group were established as heart failure mouse models through intradermal injection of isoproterenol solution (5 mg·kg¹·d⁻¹) via the dorsal subcutaneous site. Starting from the first day of modeling, corresponding doses of the drug were administered via gavage daily for 28 consecutive days. Cardiac function was monitored using echocardiography, and cardiac tissue morphology was observed through Masson staining on the day after the last time administration; transcriptomic sequencing was employed to identify differentially expressed genes across groups, with validation of mRNA and protein expression levels of key differentially expressed genes conducted throu
gh real-time quantitative PCR and Western Blotting.
Results
2
Compared with the normal group, the left ventricular ejection fraction (LVEF) and left ventricular short-axis shortening rate (LVFS) values of the model group mice decreased, while the left ventricular end diastolic diameter (LVIDd) and left ventricular end systolic dia-meter (LVIDs) values increased (
P
<0.05), the cardiac cell clearance structure was disordered, and the blue stained collagen fibers increased. Compared with the model group, echocardiographic analysis of mice in all treatment groups showed an increase in LVEF and LVFS values, and a decrease in LVIDd and LVIDs values (
P
<0.05), and the cardiomyopathological changes were improved to varying degrees. Compared with the model group, the high-dose group showed 831 upregulated genes and 391 downregulated genes. Through GO and KEGG analyses, peroxisome proliferator-activated receptor (PPAR) signaling pathway was identified as the key pathway, with ACSL1, SCD1, PPARα, Angptl4, CPT1α, and PGC-1α being the key differentially expressed genes. Compared with the normal group, the mRNA and protein levels of ACSL1, SCD1, PPARα, Angptl4, CPT1α, and PGC-1α in cardiac tissues of model group mice significantly reduced (
P
<0.05); compared with the model group, all intervention groups showed increased expression levels of these biomarkers in cardiac tissues compared to the model group (
P
<0.05). Compared to low-dose
Shenyu Yangxin
group, the mice in high-dose
Shenyu Yangxin
group demonstrated elevated mRNA and protein expression levels of ACSL1, SCD1, PPARα, CPT1α, and PGC-1α in cardiac tissues (
P
<0.05).
Conclusion
2
Shenyu Yangxin Capsules
may treat heart failure by improving myocardial fibrosis and regulating the expression of ACSL1, SCD1, PPARα, Angptl4, CPT1α, and PGC-1α genes and proteins.
