Objective To study the effect and mechanism of different combinations of sanchinoside monomers on monocyte-endotheliocyte adhesion. Methods Oxidized low density lipoprotein(ox-LDL)was added to endotheliocytes processed by sanchinoside monomers and their different combinations. Then rose bengal staining was used to detect adhesion of human THP-1 monocytes to human umbilical vein endothelial cells(HUVECs)
and enzymelinked immunosorbent assay was used to detect the expression of intercellular adhesion molecule-1(ICAM-1).Results Ox-LDL could enhance adhesion of THP-1 to HUVECs.After processed by Rg1 and Rb1
monocyte-endotheliocyte adhesion and expression of ICAM-1 decreased significantly(P<0.05)
especially in combination groups such as Rg1:Rb1
Rg1:R1
and Rb1:R1(P<0.01).Conclusion Rg1 and Rb1 can obviously inhibit moncyte-endotheliocyte adhesion induced by ox-LDL
especially in different combinations of Rg1
Rb1 and R1.Rg1
Rb1 and their combinations could inhibit monocyte-endotheliocyte adhesion by down-regulating the expression of ICAM-1
which might be related to its protection effect on endotheliocytes.
