Objective To study the syndrome character of the rats with spleen yang deficiency and spleen qi deficiency induced by reserpine.Methods Totally 80 SD rats were included in the experiment.Spleen deficiency model rats groups were made by intraperi- toneal injection of reserpine.Sijunzi Tang（Decoction of Four Noble Ingredients）and Lizhong Wan（Bolus for the Function of Middle Jiao）which were the representative medicines for spleen qi deficiency and spleen yang deficiency syndromes respectively were applied for treatment.Various indices tested were for clustering analysis.The clustering scheme was made to analyze the distinction of sample and index to study the correlation between the indices and spleen qi deficiency and spleen yang deficiency syndromes.Results Up to 12 indices have a good differential validity between the normal group and model groups:the indices of interleukin-1（IL-1）

vasoactive intestinal peptide（VIP）

and body weight of both groups had the same changing tendency;the indices of spleen T lymphocyte prolifer- ation and transforming growth factor（TGF）of Lizhong Wan group were higher than those of Sijunzi Tang group;the indices of gas- trin

and interleukin-6（IL-6）of Lizhong Wan group were lower than those of Sijunzi Tang group. Conclusion The normal group and model group have good differential validity

the spleen deficiency models were successfully made with reserpine.Effect of Lizhong Wan and Sijunzi Tang on the rats with spleen deficiency is in accord with TCM theory.
