Objective To analyze the correlation between TCM Syndromes of chronic aplastic anemia(CAA)patients and the transcription factor T-bet/GATA-3 and its related signal paths from the Th cells imbalance. Methods The CAA patients
who were diagnosed as spleen and kidney yang deficiency type and spleen and kidney yin deficiency type
20 cases each
were tested with realtime PCR to determine the expression of T-bet
GATA-3
STAT4
and STAT6 mRNA of the mononuclear cells of peripheral blood(PBMNC)
and the ratio of Th1 and Th2 and the levels of IFN-γ
IL-12
and IL-4 in the cultured supernatant of PBMNC were examined with flow cytometry(FCM)and enzyme linked immunosorbent assay(ELISA).Twenty healthy people were taken as the normal control group for comparison. Results The PBMNC T-bet
STAT4
T-bet /GATA-3
Th1
IFN-γ
and IL-12 of those with spleen and kidney yin deficiency were all higher than those of spleen and kidney yang deficiency type(P<0.05)
and the above indexes of both treated groups were all higher than the normal control group.GATA-3
STAT6
Th2
and IL-4 of the three groups were without significant difference(P>0.05).Conclusion Compared with spleen and kidney yang deficiency patients
spleen and kidney yin deficiency patients have more serious abnormal activation of IFN-γ/T-bet and IL-12/STAT4 paths and Th1 excursion
and they have also more obvious immune abnormality.
