Objective To investigate the inhibitive effect and mechanism of Nansheteng(Celastrus orbiculatus) extract on transplanted hepatic carcinoma and angio genesis in the Hepa1-6 tumor bearing mice. Methods Sixty C57BL/6 mice were randomized into normal control group
model group
cisplatin group
and Nansheteng extract large
middle
and small dosage groups
10 mice in each group.Except the normal control group
Hepa1-6 cells(1×106) 0.1ml were inoculated subcutaneously in the axilla of left anterior limb of each mouse. Two weeks later
cisplatin group was given cisplatin 1mg/kg by intraperitoneal injection
while Nansheteng large
middle
small dossage group was given Nansheteng extract of 10
20
40mg/kg respectively by gastric perfusion. All the mice were medicated for 28 days in succession and were sacrificed 24 hours after the last medication. The weights of tumors and apoptosis in the transplanted tumor tissues were measured. The expression of vascular endothelial growth factor(VEGF) in the transplanted tumor tissues was determined. Results Compared with the model group
the average weight of tumors in the Nansheteng extract large
middle and low dosage groups was reduced(P<0.05 or P<0.01).The apoptosis in the transplanted tumor tissues of the medicated groups was significantly more than that of the model group
and at the same time the VEGF expression was significantly less. Conclusion Nansheteng ethyl acetate extract is effective in inhibiting the tumor growth. Its mechanism may be related to promoting the apoptosis and inhibiting the angiogenesis of tumor.
