Objective To discuss the formation mechanism of blood-stasis syndrome in essential hypertension(EH)through the TLR-NF-κB signal transduction pathway and the effect of astragalus polysaccharide. Methods Eighty patients with EH were differentiated into blood-stasis syndrome group of 40 cases and non-blood-stasis syndrome group of 40 cases
and 30 healthy people were selected from volunteers(healthy group). The serum of each subject of all groups was collected. The real-time PCR method was adopted to measure the expression of Toll-like receptor 4(TLR4) mRNA.Different concentrations of astragalus polysaccharide(APS)were given to intervene the expression of human umbilical venous endothelial cell clone TLR4 and the activation of nuclear transcription factors-κB(NF-κB)induced by lipopolysaccharide(LPS).The PCR method was used to measure the expression of TLR4 and inhibitory κBα(I-κBa)mRNA and the immune imprinting method was applied to detect the expression of TLR4 protein and I-κBa protein. Results The expression of TLR4 mRNA in blood-stasis group and non-blood-stasis group was significantly higher than that of healthy group(P<0.01)
and it was significantly higher in the blood-stasis group than in the non-blood-stasis group(P<0.01).The expression of TLR4 mRNA in the LPS induction group was significantly higher and the expression of I-κBa mRNA was significantly lower than that in the healthy group(P<0.01).In the comparison of the APS big-
medium- and small-dose groups with the LPS induction group
the expressions of TLR4 and I-κBa mRNA were both statistically different(P<0.01).And in the comparison among the APS big-
medium- and small-dose groups
the difference of expressions of TLR4 and I-κBa mRNA was also statistically significant(P<0.01).Conclusion Hypertension patients with blood-stasis syndrome have more serious inflammation and immune disorder than those with non-blood-stasis syndrome. The inflammation and immune disorder mediated by TLR-NF-κB signal pathway is one of the mechanisms of formation of blood-stasis syndrome of EH.
