Objective To analyze the molecular mechanism of three formula of regulating and supplementing the lung and kidney for chronic obstructive pulmonary disease（COPD）.Methods THP-1 monocytes and macrophages were stimulated with cigarette smoke extract（CSE）

lipopolysaccharide（LPS）or their combination（LC）in vitro.Every kinds of stimulants were divided into normal group（20%normal rat serum was added）

stimulant group（20%normal rat serum and 5%CSE

or 75pg/ml LPS

or 5%CSE and 75pg/ml LPS were added）

supplementing the lung and kidney group

supplementing the lung and spleen group

and boosting qi enriching the kidney group（20%rat serum containing relevant formula and 5%CSE

or 75 pg/ml LPS

or 5%CSE and 75pg/ml LPS were added）.Tumor necrosis factor-α （TNF-α ）

interleukin-1β（IL-1β）

interleukin 3（IL-3）

granulocyte-macrophage colony stimulating factor（GM-CSF）

transforming growth factor-β（TGF-β）

matrix metalloproteinase-9（MMP-9）

tissue inhibitor of metalloproteinase 1 （TIMP-1）

superoxide dismutase（SOD）

CD36and phosphatidic acid phosphatase 2B（PPAP2B）were detected in all groups.The transcription factor of cytokine was analyzed with network tools.Results Comparing with the normal group

the secretion of MMP-9 was increased and the secretion of TNF-α and PPAP2B was decreased in CSE group

LPS group and LC group;the secretion of TIMP-1 was decreased in CSE group

the secretion of TGF-β was increased in LPS group and the secretion of GM-CSF was increased in LC group（P<0.05or P<0.01）.The regulation to the cytokine secretion of three formula was different（P<0.05or P<0.01）.More cytokines and transcription factors could be regulated by supplementing the lung and kidney formula.Conclusion The effect of CSE and LPS on the cytokine secretion of THP-1 cells is different.All the three formula of regulating and supplementing the lung and kidney could regulate the cytokine secretion of THP-1 cells stimulated with CSE and LPS

among which the effect of supplementing the lung and kidney formula is obvious and broad

followed by supplementing the lung and spleen formula.
