Objective To study the mechanism of Tanshinone ⅡA(TsⅡA) in promoting angiogenesis. Methods The cultured human umbilical vein endothelial cells(HUVEC-C) were randomized into the blank group
dimethyl sulfoxide(DMSO
a final concentration of 1%) group
vascular endothelial growth factor(VEGF
a final concentration of 5 μg/L) group and TsⅡA(a final concentration of 6 mg/L) group. The cell morphology was observed in each group 48h after the corresponding drugs were added. The expression of endothelial nitric oxide synthase(eNOS) and phosphoinositide 3-kinase(PI3K) was detected by Western blotting. Results The HUVEC-C cells in the VEGF group and TsⅡA group were fully shaped and significantly abundant with a narrow cell gap and no shedding.Comparing with the blank group
the expression of eNOS and PI3K in the VEGF group and TsⅡA group was significantly increased(P<0.05 or P<0.01).Conclusion TsⅡA can increase the expression of eNOS and PI3K in HUVEC-C
