Objective To observe effect and possible mechanism of action of Shen Fu Injection on the intestinal mucosal barrier function of sepsis rats. Methods 48 Sprague Dawley rats were randomly divided into control group
model group
and Shen Fu Injection group
with 16 rats in each group. The sepsis models were produced with cecum ligation perforation in all rats except those in the control group. Two hours after successful molding
Shen Fu Injection group was given 20 ml / kg of Shen Fu Injection intraperitoneally
the control group and the model group were given equivalent amount of normal saline intraperitoneally. 12 h and 24 h after the surgery 8 rats were executed respectively in each group to observe and grade the pathological changes of small intestinal mucosa. ELISA method was used to detect the level of secretory Ig A in small intestinal mucosa. Flow cytometry was applied to detect percentage changes of small intestinal mucosa CD3+and γδT cell. Results 12 h and 24 h after the surgery
compared with the control group
the score of the small intestine mucosa damage index increased significantly
but the level of secretory Ig A in small intestinal mucosa and percentages of CD3+and γδT cells decreased significantly in the model group( P < 0. 05). Compared with the model group
the score of the small intestine mucosa damage index decreased significantly
but the level of secretory Ig A in small intestinal mucosa and percentages of CD3 and γδT cells increased significantly in the Shen Fu Injection group( P < 0. 05). Conclusion Shen Fu Injection may protect the intestinal mucosal barrier function of Sepsis rats by promoting small intestinal mucosa to secrete secretory Ig A and increasing the expression of CD3+and γδT cell.
