Objective To explore the possible functional mechanism of ganoderan in improving anti-tumor immune response. Methods Melanoma cells of mice B16F10 cultured in vitro were treated using ganoderan
and were divided into low( 200 μg / ml)
middle( 400 μg / ml)
and high( 600 μg / ml) dosage groups according to the ganoderan concentration
while RPMI-640 nutrient solution was used as the control group. Mice spleen lymphocytes 1 ×106/ hole were added into ganoderan treated B16F10 cells
and after being cultured for 18 h or five days
respectively
the expression of activated molecule of CD69 and FASL on the surface of spleen lymphocytes were examined respectively. Mice spleen lymphocytes 5 × 105/ hole were added into Gl-PS treated B16F10 cells
and after being cultured for 5 days
the content of IFN-γ was detected using ELISA method. Results CD69 levels in low
middle and high dosage groups were remarkably higher than that in control group( P < 0. 05). FASL levels in middle and high groups were remarkably higher than that in control group( P < 0. 05); and FASL levels in middle and high groups were remarkably higher than that in low group( P < 0. 05). There were no statistically significant differences in CD 69 levels between each dosage group( P > 0. 05). IFN-γ level in low
middle and high dosage groups were remarkably higher than that in control group( P < 0. 05). IFN-γ level in middle dosage group was remarkably higher than that in low and high dosage groups( P < 0. 05). Conclusion Ganoderan could improve anti-tumor immune immunocompetence of lymphocytes
which may had a relationship with activating the expression of CD69 and FASL of mice spleen lymphocytes and improving secretion of IFN-γ.
