Objective To study the effect and possible mechanism of different compatibility proportion of hypaconitine
active constituent of Fuzi( radix aconiti carmichaeli) and glycyrrhetinic acid active constituent of Gancao(glycyrrhiza uralensis) on H9c2 myocardial cell with oxygen-glucose deprivation. Methods The researchers took H9c2 myocardial cell growing into compact monolayer to divide into a normal group
model group
digoxin group(80μmol /L)
hypaconitine group( 120 μmol /L)
proportion 1 ∶ 0. 5 group
1 ∶ 1 group and 1 ∶ 2 group
i. e. hypaconitine 120 μmol / L
glycyrrhetinic acid 60
120 and 240 μmol / L respectively
six holes in each group. The researchers put complete medium 1 ml into the normal group and sugar-free Earle’ s liquid 1 ml into the model group. Put the corresponding medicine into the digoxin group
the hypaconitine group and each compatibility group and used sugar-free Earle’s liquid to dilute to the corresponding concentration
making the total amount of liquid to1 ml. The other groups after putting into drug were modeled into oxygen-glucose deprivation to cultivate for 4h except the normal group. The researchers observed morphological changes of H9c2 myocardial cell in each group and detected the content of tumor necrosis factor α( TNF-α)
interleukin 1β( IL-1β) and creatine kinase( CK) and the expression of Caspase-3
Fas-L and Fas protein. Results Myocardial cells grew adhering to wall in the normal group and most of them were fusiform with irregular star and formed radiate cell clusters. In the model group
refractivity of myocardial cell body declined with the endochylema shrinking. The researchers found dying cells falling off from wall with strong refractivity. Compared with the model group
cellular form obviously improved in the each compatibility group and the most obviously in the 1 ∶ 1 group. Compared with the normal group
the content of TNF-α
IL-1β and CK and the expression of Caspase-3
Fas-L and Fas protein all increased in the model group( P < 0. 01). Compared with the model group
each index didn’t change significantly in the hypaconitine group( P > 0. 05); But the content of TNF-α
IL-1β and CK and the expression of Caspase-3
Fas-L and Fas protein decreased in varying degrees in digoxin group and the each compatibility group and the best effect was in the 1 ∶ 1 group( P < 0. 05 or P < 0. 01).Conclusion Hypaconitine combined with glycyrrhetinic acid had a protective effect on myocardial cell with oxygenglucose deprivation and 1 ∶ 1 might be the optimum. The mechanism may be related to restraining inflammation and apoptosis.
