Objective To observe possible effect and mechanism of Chinese herb extracts Peiminine on HCT-116 cells in patients with colon cancer. Methods HCT-116 cells in colon cancer patients were inoculated in 6-well plate with 1 ml in each hole
summing to 2000 cells. Those cells were divided into the control group and the Peiminine group. There were three parallel samples in each group. The Peiminine group was put Peiminine 200 μg into each hole. The final working concentration was 200 μg / ml. The control group was put isopyknic dimethyl sulfoxide.Seventy-two hours after medication
drew out medium for extracting total RNA. Gene chip technology was used to detect differential gene expression profile in the two groups and made GO
KEGG enrichment analysis and cluster analysis for differential genes. Results Totally 37 differential genes were screened out including 5 significantly upregulated genes and 32 down-regulated genes( differences multiples≥2
P < 0. 05). The main influenced access of GO analysis focused on the regulation and control of thermogene
motor behavior
uracil nucleotide metabolism process
adrenoceptor signal channel
adipose tissue metabolism and G-protein-coupled receptors signal channel in the biological process
also foucused on the activity of adrenoceptor binding and uracil nucleoside phosphorylase in the molecular function. The main influenced access of KEGG enrichment analysis focused on the drug-metabolizing enzyme
pyrimidine metabolism
synthesis and degradation of ketone body
vitamin B6 metabolism and ribosomes. The results of Cluster analysis showed the RPL and PHLDA1 of up-regulated genes contacted more closely in the Peiminine group than in the control group and the UPP1 and ARRDC3 of down-regulated genes contacted more closely. Conclusion Peiminine may obviously affect the pyrimidine metabolic pathway of HCT-116 cells of colon cancer
inhibit the proliferation of the cells and promote the energy metabolism.
