Objective To explore possible mechanism of Huwei San( 护胃散) in treating gastric mucosal injury in rush entry into plateau. Methods Randomly divided 350 Wistar rats into Beijing group
Xining group
Madoi blank group
Madoi positive control group and Huwei San high
middle and low dose groups
with 50 rats in each group. Except for Beijing group
the other 300 rats were delivered to Xining city by air. The Madoi blank group
Madoi positive control group and Huwei San high
middle and low dose groups were successively delivered to Madoi County by bus on that day. Beijing group
Xining group and the Madoi blank group were perfused with 2ml saline.Madoi positive control group was given Jinghua Weikang Capsule( 荆花胃康胶丸) 2. 4 g/( kg·d). Huwei San low
middle and high dose group were respectively given Huwei San of 2. 6 g/( kg·d)
5. 2 g/( kg·d) and 10. 4 g/( kg·d)
for 7 days totally. Ten rats in each group were sacrificed on the 1st
2nd
3rd
5th and 7th day of the experiment. Took gastric mucosal tissue and conducted pathological observation. At the same time
examined the expression of mRNA of vascular endothelial growth factor( VEGF) and basic fibroblast growth factor( bFGF) in gastric mucosa. Results There were scattered hemorrhagic spots
erosion or superficial ulcer in surface of gastric mucosa and defect change in local mucosa of rats in the Madoi blank group; congestion and inflammation exudation in submucosal vessels in the Huwei San high dose group improved much compared with the Madoi blank group. Compared with that in the Madoi blank group at the same time
there were different levels of expression increase of VEGF
and bFGF mRNA in the Huwei San high and middle dose groups on the 3rd and 5th day. And on the 2nd and 3rd day
the expression increase of VEGF mRNA in the Huwei San high dose group was superior to the Madoi positive control group( P < 0. 05 or P < 0. 01). Conclusion Huwei San could increase the expression of VEGF and bFGF mRNA in gastric mucosa of rats with rush entry into plateau. Thus it could improve repair of gastric mucosal injury in rush entry into plateau.
