Objective To explore the action mechanism of Sanqi( Radix Notoginseng) in treating atherosclerosis. Methods Based on Traditional Chinese Medicine Systematic Pharmacology( TCMSP) database
oral bioavailability( OB) ≥30%
drug likeness( DL) ≥ 0. 18
cell permeability( Caco-2) ≥-0. 4 were taken as screening condition. Combined with the literature
the effective active ingredients of Radix Notoginseng was obtained. The target of active ingredients and related diseases were extracted through PharmMapper database. All targets of atherosclerosis were collected in OMIM
TTD
Drugbank and other databases. An active "ingredients-disease-target"network map was constructed
and was verified with the selected targets. The DAVID database was used for GO analysis and KEGG pathway enrichment analysis of the key targets. Results Totally 11 active components such as ginsenoside rh2
quercetin and β-sitosterol were screened by network pharmacology to act on 92 targets and 269 related diseases.Nine targets for direct treatment of atherosclerosis were leukotriene A4 hydrolase( LTA4 H)
retinol X receptor alpha( RXRA). GO analysis showed that biological processes were closely related to inflammation
including leukotriene metabolism and synthesis
and regulation of I-κB kinase/NF-κB signaling. KEGG analysis revealed multiple functions and signaling pathways associated with atherosclerosis. Conclusion The active constituents of Radix Notoginseng can directly regulate atherosclerosis-related targets
indirectly regulate key inflammatory factors
and then regulate inflammatory factor-related NF-κB signaling pathways and TNF signaling pathways to play a role in the treatment of atherosclerosis.
