Objective To explore the mechanism of Dahuang(Rhubarb)-Huanglian(Coptis)
a drug pair in the treatment of hypertension based on network pharmacology. Methods TCMSP database was used to mine the active components of Radix et Rhizoma Rhei and Rhizoma Coptidis. UniProt database was used to correct the official gene name of the target. Through OMIM
TTD
Pharmgkb and Disgenet-Gene database
the relevant targets of hypertension were obtained. We took the intersection of rhubarb-Coptis
a drug pair on the target points
obtaining its effective target for the treatment of hypertension. The above targets were input into the String database for protein-protein interaction(PPI) network prediction
and finally the DAVID database was used for GO and KEGG enrichment analysis to reveal the potential signal pathways of rhubarb-Coptis
a drug pair for the treatment of hypertension. Results A total of 21 active ingredients of Rhubarb-Coptis
a drug pair and 67 targets for the treatment of hypertension were screened out. Through PPI network analysis
rhubarb-Coptis
a drug pair may be related to IL6
VEGFA
JUN
TNF
TP53
PTGS2
IL1β
EGF
EGFR
NOS3 and other proteins for the treatment of hypertension. GO and KEGG analysis results showed that the targets of rhubarb-Coptis
a drug pair for the treatment of hypertension were significantly enriched in HIF-1 signaling pathway
TNF signaling pathway
calcium signaling pathway
cGMP-PKG signaling and other pathways. Conclusion Rhubarb-Coptis
a drug pair has the characteristics of multiple targets
multiple pathways and multiple pathways for the treatment of hypertension. HIF-1 signaling pathway
TNF signaling pathway
and calcium signaling pathway are the main pathways for its antihypertensive effect.
