Objective To explore the target points and signaling pathways of Weichangan Pills(胃肠安丸) in treating irritable bowel syndrome(IBS)
and to explain its potential mechanism by undertaking a network pharmacology analysis. Methods Active ingredients were obtained and screened out via TCMSP and BATMAN databases in combination with text mining methods. Target genes were searched via Uniprot database. IBS-related target points were predicted and collected through GeneCards database. Cytoscape 3.7.2 software was used to establish a drug-component-target network. String database was used to construct the protein-protein interaction(PPI) network
and made for topological analysis through Cytoscape 3.7.2 software. Molecular docking verification of active ingredients and core target points were performed by Autodock vina 1.1.2 software. DAVID database was used for gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis
and then KEGG pathway target network was constructed. Results Thirty-one active ingredients of Weichangan Pills were screened out
and 102 predicted targets were obtained
with 84 common targets matched with IBS
as well as 31 core targets. There were 100 GO biological processes and 55 KEGG signal pathways
mainly enriched in dopaminergic synapses
serotonergic synapses
tumor necrosis factor signaling pathways and other related signaling pathways. Conclusion The mechanism of Weichangan Pills for treating IBS may be related to its function of regulating gastrointestinal motility and reducing visceral sensitivity.
