Objective To investigate the effect and possible mechanism of Yinxing Mihuan Oral Liquid(YMOL
银杏蜜环口服溶液) in promoting angiogenesis. Methods YMOL at a concentration of 0.6
1.2
2.4
4.8 and 9.6 mg/ml was selected
and CCK-8 method was used to detect the cell viability and to screen out the effective and safe concentration of YMOL for the following experiment
in which the mice were classified into normal group
model group
nimodipine group
and YMOL high and low dose groups. Oxygen and glucose deprivation(OGD)(4 h)/reintroduction(R)(24 h) method was used to establish bEnd.3 in vitro model of OGD/R in all groups except for the normal group. Before modelling
10μmol nimodipine was given to nimodipine group while YMOL at selected concentration was administered in high and low dose YMOL groups
respectively. The cell scratch method was used to evaluate the migration ability of endothelial cells; the Matrigel microtubule formation assay was used to detect the in vitro angiogenesis ability of endothelial cells; expression of angiogenesis-related proteins including vascular endothelial growth factor(VEGF)
CD31
angiogenin(Ang)
hypoxia-inducible factor 1α(HIF-1α)
and von Willebrand factor(vWF) were detected by western blotting. Results The concentration of YMOL at 0.6 mg/ml and 1.2 mg/ml were selected to be used in YMOL low and high dose group
respectively. Compared to the normal group
the model group had significantly decreased cell viability and cell migration
as well as increased number of nodes
intersections
meshes
total length
branch length and trunk length that form microtubules(P<0.05); there was also an upregulation of VEGF
CD31
HIF-1α and Ang expression in the model group
but it was not statistically different with the normal group(P>0.05). Compared to the model group
YMOL high dose group could improve the cell viability and the cell mobility
and remarkably up-regulate the expression of VEGF
CD31
HIF-1α
Ang and vWF(P<0.05 or P<0.01). Conclusions YMOL could promote cell proliferation of endothelial cells and increase the cell mobility in the OGD/R induced injury model by up-regulating the expression of angiogenic factors and promoting agiogenesis.
