摘要

目的探讨中药圣和散逆转胃癌耐药细胞SGC7901/VCR多药耐药的机制。方法圣和散（浓度2.5、5和10mg/ml）处理胃癌耐药细胞SGC7901/VCR

用流式细胞技术检测P-糖蛋白（P-gp）表达

免疫组织化学SABC法及图像分析技术检测B细胞淋巴瘤/白血病基因-2（Bcl-2）。结果实验组SGC7901/VCR细胞Bcl-2表达A值（121.34±0.16）明显高于对照组（104.37±0.089）（P<0.05）

Bcl-2表达减少

P-gp表达也明显降低。结论圣和散能逆转胃癌耐药细胞SGC7901/VCR多药耐药

其作用可能与调节Bcl-2和P-gp表达有关。

Abstract

Objective:To explore the mechanism of Shenghe Powder（SHP） in reversion of multidrug resistence of SGC7901/VCR cells of gastric cancer.Methods:Gastric cancer drug-resistance cells SGC7901/VCR were treated with SHP at concentrations of 2.5

5.0 and 10mg/ml.The expression of P-glycoprotein（P-gp） was detected with flowcytometry and B-cell lymphoma/leukemia-2（Bcl-2） protein was examined by immunohistochemical SABC method and imaging analysis.Results:In SHP treated group

A value of Bcl-2 in SGC7901/VCR cells was significantly higher than that in the control group（P<0.05）

and the expression of Bcl-2 and P-gp was significantly down-regulated.Conclusion:SHP can reverse the multidrug resistance of SGC7901/VCR

which is possibly related with the changes of the expressions of Bcl-2 and P-gp.
