Objective To explore the effects of Biejiajian Pills( 鳖甲煎丸
BP) on structure and function of tumor angiogenesis and tumor microenvironment. Methods Hepa1-6 mice model of subcutaneous tumor of liver cancer was established
and then the mice with successful models were randomized into a BP group
a bevacizumab group and a control group
with 15 mice in each group. The BP group was treated with BP suspension [p. o.
6 g/( kg·d) ]
the bevacizumab group was given bevacizumab injection [i. p.
5 mg/( kg·d) ]
while the control group was given the same amount of normal saline. All groups were treated for 14 days. The tumor tissue was stripped 14 days later. The tumor microvessel density
the pericyte coverage and the basal membrane area were detected by immunohistochemical staining. The vascular permeability was detected by Evans blue. The hypoxic area was detected by immunofluorescence. Results Compared with those in the control group
the microvessel density
the content of Evans blue dye and the hypoxia area decreased
while the pericyte coverage and the basement membrane area increased in the bevacizumab group and the BP group( P < 0. 05 or P < 0. 01). The Evans blue dye content and the hypoxic area in the BP group were higher than those in the bevacizumab group( P < 0. 01). There was no significant difference in the microvessel density
the pericyte coverage and the basement membrane area between the bevacizumab group and the BP group( P > 0. 05). Conclusion BP seems to have the function of normalizing the structure and the function of blood vessel in subcutaneous metastasis of Hepal-6 liver cancer in mice
which can improve the hypoxia state of tumor tissue.
